Identification of a protein mixture using mass spectrometry via nLC-MSMS analysis on the timsPro equipment (Bruker), after tryptic digestion and prior separation by reverse-phase chromatography.
The tryptic peptides initially separated by reverse phase chromatography are directly introduced into the mass spectrometer for MSMS analysis. Ion mobility will be used as an additional dimension of ion separation to increase the sensitivity of the analyses.
From the fragmentation spectra of the peptides, their amino acid sequence will be obtained and, by searching protein databases, which will lead to identification by correlation with the theoretical data, using a search engine (PEAKS, MASCOT).
