- To learn laboratory techniques and bioinformatic resources in molecular neurobiology.
- Develop skills for the use of these techniques in a practical way.
SCHEDULE OF LECTURES. (3 hours)
Session 1: Molecular Biology. Basic concepts on the basis of practices carried out.
Session 2: Marker genes for the process of neuronal differentiation. Analysis of the expression of genes using riboprobes.
Session 3: How to determine the function of a gene (I). Overexpression of genes in vivo: transgenesis, transfection, injection of mRNA.
Session 4: How to determine the function of a gene (II). Gene silencing in vitro and in vivo: morpholinos, siRNA.
INTERACTIVE TEACHING PROGRAMME (laboratory and seminar of bioinformatics). (26 hours)
Practice 1. Extraction of total brain RNA and cDNA generation.
Practice 2. Bioinformatics: searching for conserved sequences in databases and use of software for the design of primers. Amplification of cDNA by PCR.
Practice 3. Cloning of cDNA in a vector (ligation and transformation in competent cells). Obtaining plasmid DNA.
Practice 4. Obtaining antisense riboprobes for RNA detection by in situ hybridization: linearization of plasmid DNA, purification of linear DNA, transcription and purification of riboprobes. Obtaining of sense riboprobes.
The professor will provide all necessary bibliographic materials in paper or pdf format.
The student should possess and enhance the following competencies and generic skills:

-Ability for manual work in a laboratory of molecular neurobiology.
The students will be divided in pairs during the course. We will use:
-Introduction to the theoretical concepts.
-Laboratory work and seminars.
-Assistance and participation to lectures, labs and seminars is mandatory. 30% of the final qualification.
-The student must pass a final test that will represent 70% on the final qualification.
30 presential hours:
-Theoretical basis about topics related to the proposed practical activities: 3 hours.
-Development of practical activities in the laboratory: 24 hours.
-Seminar: 2 hours
-Exam: 1 hour
40 hours of student work:
-Personal work of study and consultation of bibliographical material and information obtained during the development of the activities: 40 hours.
-Assistance and active participation in the scheduled activities.
-Study and consultation of bibliographical material and information obtained during the development of the activities.
-Clarification of possible doubts with the professor.
Material that needs to be provided by the student: Lab coat, calculator, notebook.
